Here's one to have a play with.
it combines 100% K, alpha modes and blend modes, all of which would effect the 100% K
you can of course adjust the error message as well.

This is setup for fills, but a similar one should also be added for strokes.

Glad you resolved it.
However it might be time to start thinking about upgrading that version 10. It was released in 2010 so it's getting on a bit.
Windows 7 is no longer supported starting next year, and with PDF 2.0 and PDF/X-6 coming next year, it might be time to give it some thought.

You can actually go to 10 update 3

There's not many options or preferences for that. It should just load the color libraries. Anything change on the system? Are you on PitStop 10 or 10 update 1. Just another point on this. When you assign an object to a layer in PDF, it does not move the object or change the viewing order of the file. As you rightly say the problem here is the sheer diversity of different spot color names that you are going to receive. The selection of the colors is possible with a regular expression, it's the placing it on a layer of the correct name that's the tricky part. I assume this is for a proof that show...

If it's a black and white scan you could also try using the color curve function to remove anything below say 5% and making it white, that's worked for me in the past

I've played with this in the past and it can work, but it's tricky. One of the issues is if there are other objects that come into play and knockout the color. Ensuring the same knockout is there when inverted can be challenging. For instance here you have the egg knocking out. So your negative must...

Had to check myself. We added two new options, Before and After Selected pages Screen Shot 2019-09-06 at 4.16.57 PM.png That's why it now works.
